What is Kisspeptin-10?
Kisspeptin-10 is a short fragment of kisspeptin, a peptide that sits at the very top of the reproductive hormone system. Kisspeptin is essentially the master switch for the whole cascade: without it, puberty does not start. That makes kisspeptin-10 a genuinely interesting research tool, and it has been studied in real human trials for fertility and reproductive signaling, though it is not a self-use product with a validated protocol.
How Kisspeptin-10 works
Kisspeptin-10 activates the KISS1 receptor on GnRH neurons in the hypothalamus. Those neurons then release gonadotropin-releasing hormone, which tells the pituitary to secrete luteinizing hormone (LH) and follicle-stimulating hormone (FSH), which in turn drive the testes or ovaries. In other words, it acts one full step upstream of GnRH, at the top of the chain. Its effects depend on an intact hypothalamus and working GnRH neurons, which is why it stimulates the natural cascade rather than overriding it.
What the research shows
The genetics make the case powerfully: people born with loss-of-function mutations in the KISS1 or KISS1R gene fail to go through puberty, which is how researchers know this pathway is essential. In controlled human studies, intravenous kisspeptin-10 reliably raised LH and FSH in men, even at low doses. There is also a notable quirk: its effect shows sexual dimorphism, stimulating gonadotropin release in men but not in women during the follicular phase of the cycle. Beyond fertility, kisspeptin is being explored for how it influences sexual and emotional brain processing.
Dosage and how it is used
Essentially all the solid human data comes from intravenous administration in controlled research settings. There is no standardized subcutaneous self-use protocol. Treat community dosing as unvalidated.
The dose-finding work in men is specific enough to quote. George and colleagues gave healthy men intravenous boluses across a 0.01 to 3.0 mcg/kg range and found LH release peaked at 1 mcg/kg, taking LH from 4.1 to 12.4 IU/L at the 30-minute mark. Going higher backfired: 3 mcg/kg produced a smaller response than 1 mcg/kg, a ceiling effect worth knowing about before anyone assumes more is better. Continuous infusion held the effect longer, with 1.5 mcg/kg/h over 9 hours raising mean LH from 5.2 to 14.1 IU/L and lifting pulse frequency from 0.7 to 1.0 per hour, and 4 mcg/kg/h over 22.5 hours reaching 20.8 IU/L alongside a testosterone rise from 16.6 to 24.0 nmol/L.
Those are per-kilogram intravenous research doses delivered under supervision, not a protocol you can convert to a subcutaneous vial. The reason is pharmacokinetic and it is the single most useful fact on this page: kisspeptin-10 has a plasma half-life of about 4 minutes. Kisspeptin-54, the longer fragment, sits at 27.6 minutes, roughly six times longer. A 4-minute half-life is why the trials that wanted a sustained effect used infusion pumps rather than injections, and it is the honest answer to why a weekly or daily self-administered schedule has no research behind it.
Research vials are commonly sold in 5 mg and 10 mg sizes. Neither number comes from a dosing study. They are supplier packaging conventions, and a 10 mg vial is not a 10 mg dose.
Side effects and safety
In the controlled research doses studied, kisspeptin-10 has generally been well tolerated, which is part of why it is attractive as a fertility tool: it nudges the body's own axis rather than flooding it with hormones. The 2011 JCEM dose-ranging work reported no adverse events across the bolus and infusion arms. That said, the reassuring safety data comes from short, supervised, intravenous studies, not from long-term or self-administered use. Its real-world use outside research is not well characterized, and it is not an approved therapy.
Read the sample sizes before you read the safety claim. The bolus arm enrolled 6 healthy men and each infusion arm enrolled 4. Numbers that small can show that a peptide raises LH; they cannot rule out an uncommon side effect. "No adverse events in 6 men over one afternoon" and "safe" are different statements.
